The transcription factor BACH1 is a master regulator of human breast cancer metastasis. Here we use gene expression array analysis to identify and compare the genes regulated by BACH1 depletion in a metastatic human breast cancer cell line. Overall design: Total RNAs were extracted from vector control 1833 cells or 1833 cells with shBACH mRNA BACH1 depletion. Affymetrix GeneChip Human Gene 1.0 ST Arrays were performed to detail the gene expression and identify the genes regulated by BACH1in metastatic human breast cancer cells.

转录因子BACH1是人类乳腺癌转移的主调控因子。本研究借助基因表达芯片分析,在转移性人类乳腺癌细胞系中鉴定并比较了BACH1敲低所调控的基因。整体实验设计:从空载对照1833细胞以及经shBACH mRNA介导BACH1敲低的1833细胞中提取总RNA;采用Affymetrix GeneChip Human Gene 1.0 ST芯片开展检测,以全面解析该转移性人类乳腺癌细胞的基因表达谱,并鉴定受BACH1调控的基因。
