In order to add to the current knowledge on the extent to which macrolide resistance associated mutations (RAMs) circulate in Chlamydia trachomatis (CT), and whether or not RAMs are involved in cases with possible treatment failure, a cohort of women with NAAT persistence despite AZI treatment were tested with a previously developed genotypic 23S rRNA gene assay. As no macrolide RAMs were detected through Sanger sequencing, the analysis was extended with a next-generation sequencing approach, in order to facilitate detection of a heterogeneous, minor population with macrolide RAMs.
为补充当前关于大环内酯类耐药相关突变(macrolide resistance associated mutations, RAMs)在沙眼衣原体(Chlamydia trachomatis, CT)中的传播范围,以及该类突变是否与可能的治疗失败病例相关的现有认知,本研究对一组在接受阿奇霉素(AZI)治疗后核酸扩增试验(nucleic acid amplification test, NAAT)仍呈持续阳性的女性队列,采用此前开发的基因型23S rRNA基因检测方法开展检测。由于通过桑格(Sanger)测序未检测到大环内酯类耐药相关突变,研究进一步采用下一代测序方法扩展分析流程,以实现对携带大环内酯类耐药相关突变的异质性低丰度种群的检测。
