CD96+ and CD96- NK cells from human livers

To better define primary CD96+ NK cells and CD96- NK cells from human liver, we isolated primary hepatic lymphocytes from healthy liver and purified CD96+ and CD96- NK cells through negative selection and flow cytometry sorting. Purified NK cells from human liver tissues were first enriched by MACS using NK Cell Isolation Kit (Miltenyi Biotec, German) and CD96+/- hepatic NK cells were isolated by FACS Aria cell sorter (BD Biosciences, United States) to attain a purity greater than 95%. Expression profiling of CD96+ hepatic NK and CD96- hepatic NK cell types. Three healthy donors were pooled for each cell type.

为精准界定人源肝脏中的CD96阳性（CD96+）自然杀伤（NK）细胞与CD96阴性（CD96-）NK细胞，本研究从健康肝脏组织中分离原代肝淋巴细胞，并通过阴性筛选与流式细胞术分选纯化CD96+及CD96- NK细胞。首先采用德国美天旎（Miltenyi Biotec）公司的NK细胞分选试剂盒，借助磁珠分选（Magnetic Activated Cell Sorting, MACS）技术富集人肝脏组织中的NK细胞，随后使用美国BD生物科学（BD Biosciences）公司的FACS Aria细胞分选仪分离CD96+/-肝NK细胞，最终获得纯度高于95%的目标细胞群。本研究对CD96+肝NK细胞与CD96-肝NK细胞开展了表达谱分析，每种细胞类型均混合3名健康供者的样本进行实验。
