12-week-old female CAGGCre-ER-Tead1+/+ (control) and CAGGCre-ER; Tead1flox/flox (inducible global knockout; igKO) mice, and Myh6-MerCreMer-; Tead1flox/flox mice (control) and Myh6-MerCreMer+; Tead1flox/flox (inducible cardiac-specific knockout; icKO) and were subjected to IP injection of tamoxifen (50 mg/kg) for 5 consecutive days. Five days post the last injection, total RNA from the mouse hearts was extracted by TRIzol (Invitrogen). The RNA-seq library was constructed with Ribo-Zero library construction kit (illumina). RNA-sequencing was performed on a HiSeq 3000 system (Illumina) in 50-bp length at Genome Technology Access Center of Washington University.

12周龄雌性CAGGCre-ER-Tead1+/+(对照)、CAGGCre-ER; Tead1flox/flox(诱导型全身性基因敲除,igKO)小鼠,以及Myh6-MerCreMer-; Tead1flox/flox(对照)和Myh6-MerCreMer+; Tead1flox/flox(诱导型心脏特异性基因敲除,icKO)小鼠,均连续5天腹腔注射他莫昔芬(tamoxifen,50 mg/kg)。末次注射后第5天,采用TRIzol试剂(Invitrogen)提取各组小鼠心脏组织的总RNA;使用Ribo-Zero建库试剂盒(illumina)构建RNA测序(RNA-seq)文库;随后于华盛顿大学基因组技术访问中心(Genome Technology Access Center of Washington University),采用Illumina HiSeq 3000测序系统进行50bp读长的RNA测序。
