A total of 40 healthy people (20 males and 20 females) from an office located in Xiamen city, Fujian, China (24°36’ N, 118°03’E) were selected for study and swabs were taken from their keyboard, mouse, surrounding dust and all ten fingers. Dust samples from the office environment were taken as being representative of deposited aerosols. Bacterial 16S rRNA genes and fugal ITS2 regions were amplified using the primer set of 338F/806R and ITS7/ITS4, respectively. The PCR amplicons were sequenced by LC-BIO Bio-tech Ltd (Hangzhou, China), using an Illumina Miseq PE300 platform.
本研究选取位于中国福建省厦门市(北纬24°36′,东经118°03′)某办公室的40名健康志愿者(男性20名、女性20名)作为研究对象,采集其使用的键盘、鼠标、周边环境灰尘及双手十指的拭子样本。其中,办公室环境灰尘样本被用作沉积气溶胶的代表性样本。研究分别采用338F/806R引物对扩增细菌16S rRNA基因,采用ITS7/ITS4引物对扩增真菌ITS2区域。聚合酶链式反应(Polymerase Chain Reaction,PCR)扩增产物由中国杭州LC-BIO生物科技有限公司(LC-BIO Bio-tech Ltd)使用Illumina Miseq PE300平台完成测序。
