To find regulated miRNAs during peak inflammation of rheumatoid arthritis (RA), we have collected synovium from mouse STA model at day 0 (Non Arthritic) and day 10 (Peak Inflammation). For miRNA profiling, we used high-throughput BioMark Real-Time PCR system (Fluidigm, South San Francisco, CA) We analyzed expression of 750 miRNA in Non-arthritic synovium pooled from 3 mice and Arthritic synovium at peak inflammation pooled from 10 mice. Two biological replicates for each time points. Two technical replicates per sample.
为筛选类风湿关节炎(RA,rheumatoid arthritis)炎症峰值阶段的调控性微小核糖核酸(miRNA,microRNA),我们采集了小鼠STA模型在第0天(非关节炎状态)和第10天(炎症峰值阶段)的滑膜组织。为开展miRNA表达谱分析,我们采用了高通量BioMark实时荧光定量PCR系统(Fluidigm公司,美国加利福尼亚州南旧金山)。我们分别对3只小鼠混合的非关节炎滑膜组织,以及10只小鼠混合的炎症峰值阶段关节炎滑膜组织中的750种miRNA表达水平进行了分析。每个时间点设置2次生物学重复,每份样本设置2次技术重复。
