Genomewide DNA methylation profiling of saline, diesel exhaust particiles and house dust mite treated human airway epithelial cells. Methylation profiles were generated by the Illumina Infinium MethylationEPIC beadchips. Overall design: Human bronchial epithelial cells were treated with PBS, diesel exhaust particles (5ug/cm2), and house dust mite (25ug/ml) for 24 hours. Genomic DNA were extracted and treated with or without oxidation following the TrueMethyl® protocol (NuGen) followed by bisulifte conversion before being hybridized to the Illumina Infinium MethylationEPIC beadchips
本数据集针对生理盐水、柴油机尾气颗粒物及屋尘螨处理的人气道上皮细胞开展全基因组DNA甲基化谱分析。甲基化谱通过Illumina Infinium MethylationEPIC 微珠芯片(Illumina Infinium MethylationEPIC BeadChip)生成。实验设计概况:将人支气管上皮细胞分别以磷酸盐缓冲液(Phosphate Buffered Saline,PBS)、柴油机尾气颗粒物(浓度为5μg/cm²)及屋尘螨(浓度为25μg/ml)处理24小时。提取基因组DNA后,按照TrueMethyl®试剂盒(NuGen公司)的操作流程进行氧化与非氧化处理,随后进行亚硫酸氢盐转化,最后将样本与Illumina Infinium MethylationEPIC 微珠芯片进行杂交。
