Embryos were collected, aged, mock-treated/treated with 40Gy gamma radiation, and allowed to recover for 1.5 hours. Targets from 3 sets of wild type (yw, w1118) and 2 sets of mutant (Dmp53NS) biological replicates were generated and the expression profiles were determined using Affymetrix Drosophila Genechip 1 arrays. Comparisons between the sample groups allow the identification of genes with radiation-responsive and Dmp53-dependent expression patterns. 收集胚胎并按发育时长进行龄期标定,将胚胎分为假处理组与40Gyγ射线辐照处理组,随后使各组胚胎恢复培养1.5小时。针对3组野生型(Wild Type,yw、w1118)及2组突变型(Dmp53NS)生物学重复(biological replicates)样本生成检测靶标,并采用Affymetrix果蝇基因芯片1阵列(Affymetrix Drosophila Genechip 1 arrays)检测其基因表达谱(expression profiles)。通过不同样本组间的比对分析,可鉴定出具有辐射响应且依赖Dmp53的基因表达模式的基因。关键词:应激响应、基因修饰。实验设计:共分析3组配对的野生型对照与辐照样本,以及2组配对的突变型(Dmp53NS)对照与辐照样本的生物学重复。
