The opportunistic fungal pathogen Cryptococcus neoformans causes life-threatening meningitis in immunocompromised individuals. The expression of virulence factors including capsule and melanin is in part regulated by the cyclic-AMP/Protein Kinase A (PKA) signal transduction pathway. This study investigates the influence of PKA on the composition of the intracellular proteome to obtain a comprehensive understanding of the regulation that underpins virulence. PGAL7::PKA1 strains were grown under Pka1-repression (glucose-containing medium) and Pka1-induction (galactose-containing medium) conditions.
机会致病性真菌新生隐球菌(Cryptococcus neoformans)可导致免疫功能低下人群罹患致命性脑膜炎。包括荚膜与黑色素在内的毒力因子的表达,部分受环腺苷酸/蛋白激酶A(cyclic-AMP/Protein Kinase A, PKA)信号转导通路调控。本研究通过探究PKA对细胞内蛋白质组组成的影响,以全面阐明支撑病原菌毒力的调控机制。将PGAL7::PKA1菌株分别置于PKA1阻遏(含葡萄糖培养基)与PKA1诱导(含半乳糖培养基)培养条件下进行培养。
