3D打印支架的生物相容性检测数据集,使用共聚焦荧光显微镜拍摄分析支架表面活死细胞状态,并针对染色结果进行存活细胞数据统计;使用共聚焦显微镜对支架表面细胞骨架进行观察与拍摄存档,使用酶标仪测量450 nm处吸光度检测细胞活力;利用CCK8试剂盒检测灭菌后芯片材料的细胞增殖性能;在每只动物去毛的肩胛骨内侧部位皮内注射乳化剂(0.9 %氯化钠注射液和芝麻油浸提液,与弗氏完全佐剂混合)进行皮内局部诱导,按Magnusson 和Kligman 分级标准行评分,测定支架致敏性;采用0.9%(g/ml)无菌氯化钠溶液和二甲基亚砜(DMSO)分别制成了浸提液,使用平板掺入法,在有或无活化代谢系统(Aroclor1254 诱导的大鼠肝脏S9)的条件下,采用四种组氨酸缺陷型的鼠伤寒沙门氏菌(TA97、TA98、TA100 和TA1535)和大肠杆菌(WP2 uvrA)进行试验,测定支架遗传毒性、致癌性。包含测试报告,及指标对应的1项申请的发明专利(申请号:202410594798.0)。数据量约7M。
A biocompatibility test dataset for 3D-printed scaffolds. Confocal fluorescence microscopy was used to capture and analyze the live/dead cell status on the scaffold surface, and viable cell count statistics were performed based on the staining results. Confocal microscopy was also employed to observe and archive images of the cytoskeleton on the scaffold surface. A microplate reader was used to measure the absorbance at 450 nm to detect cell viability. The CCK-8 kit was utilized to test the cell proliferation performance of the sterilized chip materials. Intradermal injection of the emulsifier (a mixture of 0.9% sodium chloride injection, sesame oil extract and Freund's Complete Adjuvant) was administered on the depilated medial scapular region of each animal for local intradermal induction, followed by scoring according to the Magnusson and Kligman grading criteria to determine the sensitization potential of the scaffolds. Extracts were prepared separately using 0.9% (g/ml) sterile sodium chloride solution and dimethyl sulfoxide (DMSO). The plate incorporation method was adopted, and tests were conducted using four histidine-deficient Salmonella typhimurium strains (TA97, TA98, TA100 and TA1535) and Escherichia coli (WP2 uvrA) under conditions with or without an activated metabolic system (Aroclor1254-induced rat liver S9) to assess the genotoxicity and carcinogenicity of the scaffolds. The dataset includes test reports and one pending invention patent corresponding to the test indicators (Application No.: 202410594798.0). The total data volume is approximately 7 MB.
