We cultured differentiated hiPSC-derived cardiomyocytes either alone or in co-culture with hiPSC-derived endothelial cells to evaluate potential genes that might facilitate endothelial cell-mediated cardiomyocyte maturation. Overall design: hiPSC-derived cardiomyocytes were replated on day 10 of differentiation then cultured either alone or co-cultured with hiPSC-derived endothelial cells (ECs) for 14 days prior to sample harvest (until day 24 of cardiomyocyte differentiation; ECs were replated on day 13 of EC differentiated and harvested on dayy 27 of EC differentiation).

本研究将已分化的人类诱导多能干细胞(human induced pluripotent stem cells, hiPSC)来源心肌细胞分为单独培养组与hiPSC来源内皮细胞共培养组,以筛选可促进内皮细胞介导的心肌细胞成熟的潜在基因。整体实验设计:hiPSC来源心肌细胞于分化第10日传代铺板,随后分别在单独培养或与hiPSC来源内皮细胞(endothelial cells, ECs)共培养的条件下培育14天后收集样本(此时心肌细胞已分化至第24日;内皮细胞于其分化第13日传代铺板,并于分化第27日收获样本)。
