The mechanisms of resistance to the antibody-drug conjugate, T-DM1, were studied on clones derived from breast cancer cell line, BT474. Microarray analyses were performed to explore potential transcriptomic differences among the distinct cell lines. Overall design: RNA from parental cell line, BT474, and four resistant clones, C#1, C#2, C#3 and C#6, was purified and their gene expression profiles compared using the Affimetrix Transcriptome Analysis console. Three biological replicates were analyzed for each cell lines.
本研究以乳腺癌细胞系BT474衍生的克隆为研究对象,探究抗体偶联药物T-DM1的耐药机制。为探索不同细胞系间潜在的转录组差异,本研究采用微阵列分析技术开展相关实验。实验总体设计如下:分别纯化亲本细胞系BT474与4株耐药克隆(C#1、C#2、C#3、C#6)的RNA,利用Affimetrix转录组分析控制台(Affimetrix Transcriptome Analysis Console)对各组的基因表达谱进行比较分析;每个细胞系设置3次生物学重复。
