We report the analysis of DNA methylation in mouse chromaffin cell lines using reduced representation bisulfite sequencing (RRBS). We compared DNA methylation profiles of cell lines with or without a knock-out of Sdhb gene, showing that Sdhb disruption results in a hypermethylator phenotype. Overall design: Reduced representation bisulfite sequencing of 4 mouse chromaffin cell samples (2 Sdhb wild-type and 2 Sdhb knock-out).
本研究报道了采用简化代表性亚硫酸氢盐测序(reduced representation bisulfite sequencing, RRBS)分析小鼠嗜铬细胞系DNA甲基化的相关工作。本研究对比了存在Sdhb基因敲除与未敲除Sdhb基因的细胞系的DNA甲基化谱,结果显示Sdhb基因缺失会诱导产生高甲基化表型。实验设计:对4份小鼠嗜铬细胞样本开展简化代表性亚硫酸氢盐测序,其中2份为Sdhb野生型样本,2份为Sdhb基因敲除型样本。
