In order to explore the physiological changes and molecular responses of skeletonema costatum in a diel cycle, we conducted an outdoor culture experiment under natural light in july 2019. The cells of Skeletonema costatum were placed into each of three replicate transparent plastic containers containing 40L filtered seawater. Then they were suspended in an outdoor seawater pond at Muping Coastal Station of the Chinese Academy of Sciences, China. The experiment lasted a diel cycle from 16:00 to 16:00 on the next day.

为探究中肋骨条藻（Skeletonema costatum）在昼夜周期内的生理变化与分子响应，本研究于2019年7月开展了自然光条件下的室外培养实验。将中肋骨条藻细胞接种至3个生物学重复的透明塑料容器中，每个容器盛有40升过滤海水。随后将这些容器安置于中国科学院牟平海岸站的室外海水池塘内进行培养。本次实验的周期为一个完整的昼夜周期，即从当日16:00延续至次日16:00。
