The aim of this study was to identify the biological mechanisms involved in the effects of natural compounds such as heparin analogues obtained from marine invertebrates in wound healing We performed the obtaining, purifying and chemical characterization of the dermatan sulfate extracted from Styela plicata, an ascidian species. DS was used in wound healing in vitro assay and was analyzed for its effect in the toxicity, proliferation and cellular migration of murine fibroblasts. In addition, a gene expression test was performed to determine which genes responsible for the healing process were being modulated. Overall design: All experiments was performed in biological triplicates (control group - untreated, and test group - DS treated).
本研究旨在明确从海洋无脊椎动物中提取的类肝素等天然化合物在伤口愈合过程中所涉及的生物学机制。我们完成了从被囊动物柄海鞘(Styela plicata)中提取的硫酸皮肤素(dermatan sulfate,DS)的制备、纯化与化学表征工作。将硫酸皮肤素应用于体外伤口愈合实验,并分析其对小鼠成纤维细胞的毒性、增殖及细胞迁移的影响。此外,还开展了基因表达检测,以确定调控伤口愈合进程的相关基因。整体实验设计:所有实验均设置三次生物学重复,分为对照组(未处理组)与实验组(硫酸皮肤素处理组)。
