With previously optimized conditions, the linearity range for HDAC11 was obtained and the Km for the substrate was calculated. <strong>Note: </strong>1. In the assay buffer, BSA conc. is 0.5 mg/ml (instead of 0.5%). 2. In the 7.5 ul developer solution, 40 uM of TSA (Trichostatin A) is also included.
基于此前优化的实验条件,我们获取了组蛋白去乙酰化酶11(HDAC11)的线性检测范围,并计算得到其对应底物的米氏常数(Km)。 注意: 1. 本次测定所用的缓冲液中,牛血清白蛋白(BSA, Bovine Serum Albumin)的浓度为0.5 mg/ml(而非0.5%)。 2. 在7.5 μL的显色液中,还添加了40 μM的曲古抑菌素A(TSA, Trichostatin A)。
