Angiotensin II (Ang II) mediated signaling plays a key role in the development of hypertension associated target organ damages. However, the gene expression changes regulated by Ang II in the early stage of acute cerebral, cardiac, renal, vascular injury remain unclear. we investigated Ang II–mediated gene expression alteration associated with the development of early cerebral, cardiac, renal, vascular injury by microarray assay in a mouse model. All mice were euthanized by an overdose of pentobarbital on days 1, 3 and 7 of Angiotensin II treatment. Total RNA was isolated with TRIzol (Invitrogen) from brains, hearts, kidneys and vessels (n=1-3 per group) at each time point according to manufacturer’s instructions. Gene expression profiling was performed using Affymetrix GeneChip mouse Genome 430 2.0 array according to the manufacturer’s instructions (Affymetrix, Inc., Santa Clara, CA). On the GeneChip Mouse Genome 430 2.0 Array, over 45,000 probe sets analyze the expression level of over 39,000 transcripts and variants from over 34,000 well characterized mouse genes.

血管紧张素II(Angiotensin II, Ang II)介导的信号通路在高血压相关靶器官损伤的发生发展中发挥关键作用。然而,目前对于血管紧张素II在急性脑、心、肾、血管损伤早期阶段所调控的基因表达变化仍不甚明确。本研究通过小鼠模型结合微阵列检测技术,探究了与早期脑、心、肾、血管损伤进展相关的血管紧张素II介导的基因表达改变。所有小鼠在接受血管紧张素II给药后的第1、3、7天,通过过量戊巴比妥实施安乐死。按照生产商说明书的操作步骤,于各时间点从脑组织、心脏、肾脏及血管组织中提取总RNA(使用Invitrogen公司的TRIzol试剂),每组样本量为1~3例。随后依照操作手册(美国加利福尼亚州圣克拉拉市Affymetrix公司)的规范,采用Affymetrix GeneChip小鼠基因组430 2.0芯片完成基因表达谱分析。该GeneChip小鼠基因组430 2.0芯片搭载超过45000个探针组,可对超过34000个已充分注释的小鼠基因对应的39000余种转录本及其变异体的表达水平进行检测。
