To investigate the similarity of toll-like receptor tolerance in macrophages stimulated with different toll-like receptor ligands we stimulated naïve or tolerant macrophages with ligands for TLR4, TLR2, TLR3 and TLR9. The data identifies a core set of genes that are tolerised by all ligands and genes that show TLR specific patterns. Overall design: Bone marrow derived macrophages were stimulated with four different toll-like receptor ligands; LPS, CpG, Pam3csk4 and PolyI:C. In addition some cells received a pre-treatment with the same ligand prior to restimulation to induce a tolerant state. Gene expression was measured at 4 hours post stimulation with each ligand. Three independent experiments were performed to measure gene expression changes between each macrophage group. Of the 27 samples, LPSTolerance_3 and Pam3csk4_2 were excluded from the analysis entirely due to low sample quality – they are included in this dataset for completeness only.
