To investigate early transcriptional changes upon HDACi treatment, a well characterized and p53-deficient CTCL cell line (H9) and primary malignant T cells isolated from the peripheral blood of Sezary Syndrome (SS) patients (n=3) were cultured for 6 hours in the presence of vehicle control, romidepsin (romi) or belinostat (beli) and transcriptionally profiled by RNA-seq Overall design: Comparative gene expression profiling analysis of RNA-seq data for cell lines and patients in the presence of belinostat romidepsin.
为探究组蛋白去乙酰化酶抑制剂(HDACi)处理后的早期转录组变化,本研究选取一株经充分表征且p53缺陷的皮肤T细胞淋巴瘤(CTCL)细胞系H9,以及从3例塞扎里综合征(SS)患者外周血中分离得到的原代恶性T细胞,将其分别置于溶剂对照、罗米地辛(romi)或贝利司他(beli)的培养体系中培养6小时,随后通过RNA测序(RNA-seq)完成转录谱分析。
整体实验设计:针对溶剂对照、罗米地辛或贝利司他处理下的细胞系与患者来源样本的RNA测序数据,开展比较基因表达谱分析。
