RNA-sequencing analysis of CDK12 overexpressiong ZR-75-30 cell lines. Cyclin dependent kinase 12 (CDK12) is amplified in approximately 70-90% of HER2 amplified breast cancer. Results provide insight into the targets of CDK12 in HER2 positive breast cancer. Overall design: Total RNA was isolated from ZR-75-30 cell infected with control (pLVX) or CDK12 (pLVX-CDK12) lentiviruses using Trizol reagent. For each of the 2 conditions, 3 biological replicates were included. In total, 6 RNA-sequencin samples were analyzed; 3 controls and 3 CDK12 overexpressing cells.
CDK12过表达ZR-75-30细胞系的RNA测序(RNA-sequencing)分析。细胞周期蛋白依赖性激酶12(Cyclin dependent kinase 12)在约70%-90%的HER2扩增型乳腺癌中存在扩增现象。本研究结果可为解析HER2阳性乳腺癌中CDK12的调控靶点提供参考依据。实验设计:采用Trizol试剂从感染对照慢病毒(pLVX)或CDK12过表达慢病毒(pLVX-CDK12)的ZR-75-30细胞中提取总RNA。两种实验条件各设置3个生物学重复,共计6个RNA测序样本用于分析,其中3份为对照组样本,3份为CDK12过表达细胞样本。
