In this study, we have characterized various developmental stages of the free-living nematode Rhabditophanes sp. kr3021. To test the dauer hypothesis which states that the infective larval stage in parasitic nematodes evolved from the dauer stage in free-living nematodes, we compared transcriptomic profiles of the parasitic Strongloides nematodes with the RNA-seq data of the most closely related free-living species Rhabditophanes. We had previously generated developmental transcriptomes of Rhabditophanes sp. kr3021 which were submitted as ENA project ID PRJEB36286 (ERP119451), but some replicates of these previously sequenced transcriptomes showed a relatively high degree of variability. Therefore, we repeated these experiments in a more controlled setting with more replicates.
