Targeting Ewing sarcoma cells via small molecule YK-4-279 treatment and downregulation of DHX9 helicase [1]

To investigate the function of DHX9 in Ewing sarcoma cell growth and proliferation, we targeted DHX9 gene by shRNA or EWS_FLI1/DHX9 interaction by YK-4-279 treatment. Overall design: We performed gene expression profiling analysis using data obtained from RNA-seq of TC-71 cells treated with either DMSO or YK-4-279 2mM, or transfected with either control or DHX9 shRNAs.

为探究DHX9在尤因肉瘤（Ewing sarcoma）细胞生长与增殖中的功能，我们分别通过短发夹RNA（short hairpin RNA, shRNA）靶向DHX9基因，或通过YK-4-279处理干预EWS_FLI1与DHX9的相互作用。实验设计概述：我们对经二甲基亚砜（dimethyl sulfoxide, DMSO）、2mM YK-4-279处理的TC-71细胞，以及转染对照短发夹RNA或DHX9 shRNAs的TC-71细胞的RNA-seq数据开展了基因表达谱分析。
