Wnt regulates various cell responses. In dental pulp cells, Wnt signaling control cell proliferation, apoptosis, migration and differentiation. Here, the differential gene expression of human dental pulp stem cells treated with Wnt ligands or Wnt agonist was examined using a high throughput RNA sequencing technique. Results demonstrated that Wnt ligands or Wnt agonist altered numerous gene expression in human dental pulp stem cells. Cells were treated with Wnt2B (500 ng/mL) or Tideglusib (200 nM) or IWP (25 uM) for 24 h in normal growth medium. Total RNA was isolated and subjected to RNA sequencing.

Wnt信号通路可调控多种细胞应答反应。在牙髓细胞(dental pulp cells)中,Wnt信号通路调控细胞增殖、凋亡、迁移与分化进程。本研究采用高通量RNA测序(high throughput RNA sequencing)技术,对经Wnt配体(Wnt ligands)或Wnt激动剂(Wnt agonist)处理的人牙髓干细胞(human dental pulp stem cells)的差异基因表达(differential gene expression)情况进行了检测。结果表明,Wnt配体或Wnt激动剂可改变人牙髓干细胞内大量基因的表达水平。实验中,细胞于正常生长培养基中分别经Wnt2B(500 ng/mL)、替度鲁布(Tideglusib,200 nM)或IWP(25 μM)处理24小时,随后提取总RNA(Total RNA)并进行RNA测序。
