A systematic RNA-seq study comparing the transcriptional response to three independent bulky DNA damage inducing agents: UV, the chemotherapy cisplatin and benzo[a]pyrene, a component of cigarette smoke. It was performed in 2 different cell lines - GM12878 immortalized lymphoblast cell line and A549 lung cancer cell line. We performed RNA-Seq in untreated GM12878 and A549 cells, as well as 6h after being treated with 5μM of BPDE, 200μM of cisplatin, 20J/m2 of UVC. For each condition there were 3 replicates: A549 control, A549 + cisplatin, A549 + BPDE , A549 + UV , GM12878 control, GM12878 + cisplatin, GM12878 + BPDE , GM12878 + UV.
本研究为系统性RNA测序(RNA-seq)研究,旨在对比三种独立的大块DNA损伤诱导剂——紫外线(UV)、化疗药物顺铂(cisplatin)以及香烟烟雾组分苯并[a]芘(benzo[a]pyrene)所诱导的转录响应差异。本实验采用两种不同细胞系完成:永生化淋巴母细胞系GM12878与肺癌细胞系A549。我们对未处理的GM12878、A549细胞,以及分别经5μM BPDE、200μM顺铂、20J/m²短波紫外线(UVC)处理6小时后的上述细胞进行了RNA-seq测序。每个实验条件均设置3次生物学重复,具体分组如下:A549对照组、A549+顺铂组、A549+BPDE组、A549+UV组、GM12878对照组、GM12878+顺铂组、GM12878+BPDE组、GM12878+UV组。
