A murine model that mimic the decidualization and regression observed in human was used to investigate the molecular mechanisms underlying the dynamic processes in endometrium. Ovariectomized mice were treated sequentially with steroid hormones and then, to induce decidualization, oil was injected into the uterine lumen. A process similar to menstruation was induced by hormone-withdrawal. The uterine tissues were collected at 4 time-points after the induction of decidualization. Keywords: decidualization, menstruation, progesterone-withdrawal, time-course The uterine tissues were collected at 36 (T1), 48 (T2), 60 (T3) and 84 hours (T4) after the last progesterone injection. There are 4 experimental animals for each time-point, and the RNAs collected from animals within the same time-point group were pooled together for Affymetrix microarray analysis using U74Av2 Chips.

本研究采用模拟人类蜕膜化(decidualization)与退化过程的小鼠模型,探究子宫内膜动态变化背后的分子机制。对去卵巢小鼠依次给予类固醇激素处理,随后向其子宫腔内注入油剂以诱导蜕膜化。通过激素撤除(hormone-withdrawal)诱导出类似人类月经的生理过程。在蜕膜化诱导后的4个时间点收集子宫组织。关键词:蜕膜化、月经、孕激素撤除(progesterone-withdrawal)、时间进程(time-course)。具体采集时间为末次孕激素注射后36小时(T1)、48小时(T2)、60小时(T3)及84小时(T4)。每个时间点设置4只实验动物,将同一时间点组内动物提取的RNA混合后,采用Affymetrix U74Av2芯片开展微阵列分析。
