The genome of one representative taro landrace was subjected to two sequencing strategies, Oxford Nanopore Technologies and 10x Genomics linked-reads. The genome was assembled using a merged approach. Genotypic data from a mapping population (parents '230' and '255'; 96 progeny '1025') that showed resistance to Taro Leaf Blight (BioProject PRJNA381383, SRR5468959) were applied to construct a linkage map and to perform QTLs analysis to identify SNPs associated with Taro Leaf Blight resistance.
本研究针对一份代表性芋地方品种的基因组,采用牛津纳米孔测序技术(Oxford Nanopore Technologies)与10x Genomics连锁读长测序(10x Genomics linked-reads)两种策略进行测序。该基因组通过融合组装策略完成组装。利用对芋叶斑病(Taro Leaf Blight)具有抗性的作图群体(亲本为‘230’与‘255’,包含96个后代株系‘1025’,该群体的相关BioProject编号为PRJNA381383,测序数据编号为SRR5468959)的基因型数据,构建连锁图谱并开展数量性状位点(Quantitative Trait Locus, QTL)分析,以鉴定与芋叶斑病抗性相关的单核苷酸多态性(Single Nucleotide Polymorphisms, SNPs)。
