Nucleoid structuring protein H-NS silences the expression of Salmonella Pathogenicity Island 1 (SPI-1) by binding to the promoter region of SPI-1's master regulator. The experiment aims at testing whether transcription of sequences more than one Kb upstream of the regulator gene causes H-NS:DNA complexes throughout SPI-1 (and elsewhere) to disassemble (as suggested by functional assays).

类核结构蛋白H-NS通过结合沙门氏菌致病岛1(Salmonella Pathogenicity Island 1,SPI-1)主调控因子的启动子区域,沉默该致病岛的基因表达。本实验旨在验证:当调控基因上游超过1千碱基对的序列发生转录时,是否会致使分布于沙门氏菌致病岛1(SPI-1)及其他区域的H-NS-DNA复合物发生解离,该推论源于功能实验结果。
