Identification and quantification of cholangiocarcinoma (CCA) marker proteins from bile extracellular vesicles for non-invasive diagnosis

Extracellular vesicles (EVs) including exosomes are secreted into extracellular fluids, which are found in most body fluids. Since EVs contain a number of biologically active molecules such as proteins and mRNAs, there has been increasing interest in EVs as a carrier of potential biomarkers for various diseases. We previously reported that chelating reagent enhanced the recovery rate of EVs from serum, plasma, and urine. In this study, we used bile EVs which packaged biomarker molecules secreted from CCA lesion. Then we performed proteomic analysis of EVs isolated from human bile in CCA and choledocholithiasis (biliary stone) for CCA biomarkers discovery.
细胞外囊泡(Extracellular vesicles, EVs,包括外泌体(exosomes))可分泌至细胞外液中,且在绝大多数体液内均可被检测到。由于EVs携带有蛋白质、信使RNA(mRNA)等多种生物活性分子,其作为多种疾病潜在生物标志物载体的研究价值日益受到学界关注。我们此前的研究表明,螯合剂可提升从血清、血浆及尿液中回收EVs的回收率。本研究采用了包裹胆管癌(Cholangiocarcinoma, CCA)病灶分泌的生物标志物分子的胆汁EVs;随后对胆管癌及胆管结石病(胆道结石)患者的胆汁中分离得到的EVs开展蛋白质组学分析,以挖掘胆管癌相关生物标志物。
