We measured chromatin accessibility in neuronal nuclei from cortex derived from PD (18 male, 10 female) and healthy control (2 male, 2 female) brains. Each brain sample consisted of cortex from both right and left hemispheres. Th side of PD onset, age, duration of disease, as well as technical variables were used to define linear models related to DiffBind scores. Significant differences due to age, disease, sex, hemisphere, side of PD onset, or combinations were determined Overall design: Frozen prefrontal cortex was prepared and sorted to isolate neuronal nuclei. DNA was preparred for bulk ATAC-sequencing. Peaks were called with MACS2 and compared using DiffBind for differences.

本研究针对帕金森病(Parkinson's Disease, PD)患者(18名男性、10名女性)与健康对照个体(2名男性、2名女性)的大脑皮层神经元细胞核,开展染色质开放程度检测。每一份脑样本均包含左右双侧大脑皮层组织。本研究以帕金森病发病侧、年龄、病程时长及各类技术变量为自变量,构建与DiffBind评分相关的线性模型,并鉴定出由年龄、疾病状态、性别、大脑半球、帕金森病发病侧或上述因素联合作用导致的显著性差异。实验设计:制备冻存前额叶皮层样本并分选以分离神经元细胞核,提取DNA进行批量ATAC测序(bulk ATAC-sequencing);通过MACS2软件识别染色质开放峰,并利用DiffBind工具开展差异分析。
