Changes in gene expression induced by FSH in cumulus cells treated with adenosine using Agilent EmbryoGENE microarray slides. After culture, COC were were washed and cumulus cells were used for microarray analysis Overall design: Four pools of cumulus cells treated by FSH were hybridized against four pools of control cumulus cells in a dye-swap design.
本研究旨在探究经腺苷(adenosine)处理的卵丘细胞(cumulus cells)中,由促卵泡激素(Follicle-Stimulating Hormone,FSH)诱导的基因表达变化,实验采用安捷伦EmbryoGENE微阵列芯片(Agilent EmbryoGENE microarray slides)完成检测。实验对象为牛卵丘-卵母细胞复合体(cumulus-oocyte complexes,COCs),在添加腺苷的培养液中体外培养6小时。将施加FSH处理的实验组与未接受任何处理的卵丘-卵母细胞复合体对照组进行FSH生物学效应对比。培养结束后,对卵丘-卵母细胞复合体进行洗涤,分离得到的卵丘细胞用于后续微阵列分析。总体实验设计:采用染料交换(dye-swap)设计,将4份经FSH处理的卵丘细胞混合样本与4份对照卵丘细胞混合样本进行杂交。
