Stromal cells in bone are considered as a heterogenous population that have different locations and functions, though all of them can self-renew and differentiate into osteoblasts. We used single cell RNA sequencing(scRNA-seq) to dissect the heterogeneity of Stromal cell in the bone. Overall design: Stromal cell in bone marrow and bone periosteum were digested and isolated by Fluorescence-activated cell sorting (FACS). Then cells were analyzed using scRNA-seq.
骨基质细胞(Stromal cells)被认为是一类具有各异组织定位与生物学功能的异质性细胞群体,尽管所有此类细胞均具有自我更新能力,且可分化为成骨细胞。本研究采用单细胞RNA测序(single cell RNA sequencing, scRNA-seq)技术解析骨基质细胞的异质性。实验设计:对骨髓与骨膜中的基质细胞进行消化处理,通过荧光激活细胞分选(Fluorescence-activated cell sorting, FACS)分离得到目标细胞,随后对分离获得的细胞开展scRNA-seq分析。
