We used single cell RNA sequencing (scRNA-seq) to analyze the gene expression dynamics of germ cell development during spermatogenesis in the testis. Overall design: Testicular cells were isolated from mouse seminferous tubules using 2 step enzymatic digestion followed by filteration using 100 µm and 40 µm cell strainer and analyzed using scRNAseq.
本研究采用单细胞RNA测序(single cell RNA sequencing, scRNA-seq)技术,解析小鼠睾丸精子发生过程中生殖细胞发育的基因表达动态变化。实验设计概述:从小鼠生精小管中分离睾丸细胞,先通过两步酶解法完成细胞消化,随后使用100 µm与40 µm孔径的细胞筛进行过滤富集,最终采用scRNA-seq技术完成测序分析。
