To study the gene expression of wild-type L. longbeachae and a mutant in the capsule transporter gene ctrC, we grew each strain to exponential, post-exponential or stationary phase and extracted RNA for RNAseq. L. longbeachae wild-type and ΔctrC were grown in rich yeast medium (BYE) to reach exponential, post-exponential or stationary phase. RNA was isolated from quadruplicate samples and analyzed by RNAseq. Gene expression was compared for wild-type samples between exponential and post-exponential phase, and between wild-type and ΔctrC for exponential and post-exponential phase, respectively.
为探究野生型长滩军团菌(L. longbeachae)与荚膜转运蛋白基因ctrC突变株的基因表达特征,我们将各菌株培养至指数生长期、指数后期或稳定期,随后提取RNA用于RNA测序(RNAseq)。本研究中,野生型长滩军团菌与ΔctrC突变株均在富营养酵母培养基(BYE)中培养至上述三个生长阶段。从四份重复样本中分离总RNA,并通过RNAseq完成测序分析。本研究的对比分析分为两组:其一为野生型菌株在指数生长期与指数后期的基因表达差异;其二为在指数生长期及指数后期,野生型菌株与ΔctrC突变株的基因表达差异。
