Our aim was to identify the genome-wide direct LET-418 binding sites in early C.elegans embryos. a FLAG-tagged version of LET-418 was expressed in let-418(ts) mutants synchronised as early mixed stage embryos, and processed for ChIP followed by deep-sequencing. The obtained binding profile was compared to non-enriched input DNA and significant binding sites were identified.
本研究旨在鉴定秀丽隐杆线虫(Caenorhabditis elegans)早期胚胎中全基因组范围内的LET-418直接结合位点。我们将带有FLAG标签(FLAG-tag)的LET-418重组蛋白在同步化的早期混合阶段let-418(ts)突变体胚胎中表达,随后进行染色质免疫共沉淀(Chromatin Immunoprecipitation,ChIP)并结合深度测序进行分析。将获得的结合图谱与未富集的输入DNA进行比对,最终鉴定得到显著结合位点。
