Male KO (Syt10-Cre/Cre x Bmal1-flox/-) and CON (Syt10-Cre/Cre x Bmal1-+/-) mice were entrained to 12h:12h light:dark conditions for two weeks and sacrificed at 4 different time points (CT1, 7, 13, 19) on the second day after release into constant darkness conditions. Total RNA was isolated from epididymal white adipose tissue biopsies and subjected to microarray hybridization. Overall design: 3 mice were used at each time point, for both CON and MUT groups
雄性基因敲除(Knockout, KO)小鼠(基因型为Syt10-Cre/Cre × Bmal1-flox/-)与对照(Control, CON)小鼠(基因型为Syt10-Cre/Cre × Bmal1-+/-)先在12小时光照:12小时黑暗的光暗循环条件下适应性饲养两周,待其适应后,在转入恒暗环境的次日,于4个不同昼夜时间点(CT1、7、13、19)处死小鼠。从附睾白色脂肪组织活检样本中提取总RNA,随后进行基因芯片杂交检测。本实验总体设计为:对照组与基因敲除组的每个时间点均使用3只小鼠。
