3D constrained conditions promoted H3K4 methylation in ALL cells that altered the global chromatin configuration and transcriptional changes related to cell cycle and DNA replication. Overall design: CCRF-CEM cells (T-ALL cell line) were used for RNA extraction and hybridization on Affymetrix microarrays. CCRF-CEM cells were cultured in suspension or 3D conditions (a type I collagen matrix of 1,7 mg/mL) for 1 hour. Cells were lised and mRNA was extracted to compare expression profiles. We used 2 replicates of each sample.

三维约束培养条件可促进急性淋巴细胞白血病(Acute Lymphoblastic Leukemia, ALL)细胞中的H3K4甲基化,进而改变细胞全局染色质构象并引发与细胞周期、DNA复制相关的转录组改变。实验设计:本研究采用CCRF-CEM细胞(T-ALL细胞系)进行RNA提取,并在Affymetrix微阵列芯片上完成杂交反应。将CCRF-CEM细胞分别置于悬浮培养体系与三维培养体系(1.7 mg/mL的I型胶原基质)中培养1小时,收集细胞并裂解后提取mRNA,以对比两组的基因表达谱。每个样本设置2个生物学重复。
