Transcriptome is used to determine the induction response of <i>Pinellia ternata (Thunb.) Breit</i> T2 plus line (abbreviated as <i>PT2P</i> line) infected with <i>Pectobacterium carotovorum</i>. The main objective of the study was to deal with the transcriptome database of <i>PT2P</i> line resistance to soft rot pathogens to provide a new perspective for identifying the resistance-related genes and understanding the molecular mechanism. Results indicated that water soaking and tissue collapse started at 20 h after <i>PT2P</i> line was infected by <i>P. carotovorum</i>. A total of 1360 and 5768 differentially expressed genes (DEGs) were identified at 0 h and 20 h, respectively. After 20 h of infection, growth and development-related pathways were inhibited. Meanwhile, DEGs were promoted the colonization of <i>P. carotovorum</i> pathogens in specific cell wall modification processes at the early infected stage. A shift to a defensive response was triggered at 0 h. A large number of DEGs were mainly up-controlled at 20 h and were substantially used in the pathogen recognition and the introduction of signal transformation cascades, secondary metabolites biosynthesis, pathogenic proteins activation, transcription aspects and numerous transporters. Furthermore, our data provided novel insights into the transcript reprogramming of <i>PT2P</i> line in response to <i>P. carotovorum</i> infestation.

Breit)T2加系(缩写为PT2P系)感染胡萝卜软腐果胶杆菌(Pectobacterium carotovorum)后的诱导响应。本研究的核心目标为构建PT2P品系抗软腐病菌的转录组数据库,为挖掘抗病相关基因、解析其分子作用机制提供全新研究视角。研究结果显示,PT2P系感染胡萝卜软腐果胶杆菌后20小时,开始出现水浸状病斑与组织腐烂。分别在接种后0小时与20小时鉴定出1360个和5768个差异表达基因(differentially expressed genes, DEGs)。接种20小时后,与生长发育相关的通路受到显著抑制;而在侵染早期,特定细胞壁修饰过程中的差异表达基因促进了胡萝卜软腐果胶杆菌的定殖。接种后0小时即触发了防御反应的转变,20小时时大量差异表达基因呈上调表达状态,显著富集于病原体识别、信号转导级联反应、次生代谢产物生物合成、致病蛋白活化、转录调控及多种转运蛋白相关通路。此外,本研究数据为解析PT2P品系响应胡萝卜软腐果胶杆菌侵染的转录重编程过程提供了全新见解。
