Samples (2-10 mg) were extracted with 2:5:2 CDCl3:CD3OD:H2O, ground, and centrifuged to remove particulate.<br>Extracts (10 uL) were analyzed with HILIC-LC-MS using an Agilent Poroshell 120 HILIC-Z column (2.1 x 100 mm, 1.9 um particle size) at 35 C. Solvent A: 95/5 water/acetonitrile with 0.2% acetic acid and 5 mM ammonium acetate; Solvent B: 95/5 acetonitrile/water with 0.2% acetic acid and 5 m M ammonium acetate. <br>Gradient (0.3 mL/min):99% B for 1 min, decrease to 40% B over 9 min, increase to 99% B over 6 min.<br>
样品(2~10 mg)经研磨后,采用体积比为2:5:2的氘代氯仿(CDCl3)、氘代甲醇(CD3OD)与水(H2O)的混合溶剂进行提取,随后通过离心去除固相颗粒物。
取10 μL提取物采用亲水作用色谱-质谱联用法(HILIC-LC-MS)进行分析,色谱柱使用安捷伦(Agilent) Poroshell 120 HILIC-Z型色谱柱(规格2.1×100 mm,填料粒径1.9 μm),柱温设置为35℃。流动相A:含0.2%乙酸及5 mM乙酸铵的体积比为95:5的水-乙腈混合溶液;流动相B:含0.2%乙酸及5 mM乙酸铵的体积比为95:5的乙腈-水混合溶液。
梯度洗脱程序(流速0.3 mL/min):初始阶段以99%流动相B运行1 min,随后9 min内将流动相B比例降至40%,再于6 min内回升至99%。
