The goal of this study was to identify transcriptional differences between varying combinations of Tet deletion clones following six days of LIF withdrawal. These libraries were generated from cells under normal culture conditions. Overall design: RNA-seq libraries were generated for 3 WT, 3 Tet1-/-, 2 Tet2-/-, DKO, and TKO clones. Sequencing was done on a Illumina NextSeq 500 for all paired end reads

本研究旨在探究撤除白血病抑制因子(Leukemia Inhibitory Factor, LIF)并培养六天后,不同组合的Tet基因敲除克隆之间的转录组差异。本研究的测序文库均由正常培养条件下的细胞制备获得。 实验设计概述:共为3份野生型(Wild Type, WT)克隆、3份Tet1基因纯合缺失(Tet1-/-)克隆、2份Tet2基因纯合缺失(Tet2-/-)克隆、双敲除(Double Knockout, DKO)克隆以及三敲除(Triple Knockout, TKO)克隆构建了RNA测序(RNA-seq)文库。所有样本均采用Illumina NextSeq 500测序平台完成双端测序。
