Genome-wide transcriptional responses of Streptococcus gordonii Challis to AFYWWFY heptamer.. Streptococcus gordonii

Transcriptome profiling of mid-log S. gordonii cells treated for 30 minutes with 1 µM of AFYWWFY heptamer dissolved in dimethyl sulfoxide (DMSO). Overall design: One-condition experiment. S. gordonii were grown statically in 10 ml volumes to mid-log phase at 360C in 5% CO2 and then treated with either heptamer or solvent-only control and incubated for an additional 30 minutes before harvesting for RNA preparation. Experiment was repeated in different media: Todd Hewitt (TH; Difco) and chemically defined FMC medium (Terleckyj, B., Willett, N., P., and G. D. Shockman. 1975. Growth of Several Cariogenic Strains of Oral Streptococci in a Chemically Defined Medium. Infect Immun. 11(4):649–655). Biological replicates in TH: 1 control, 1 treated, 6 replicates per array. Biological replicates in defined medium: 2 control, 2 treated, 6 replicates per array. Dye swaps were performed for each sample set.

本数据集为经溶解于二甲基亚砜（DMSO）的1 μM AFYWWFY七肽处理30分钟的对数中期戈登链球菌（Streptococcus gordonii，简称S. gordonii）细胞的转录组谱分析数据。
实验设计：单条件实验。将S. gordonii以10 mL体积静置培养，于36 ℃、5% CO₂环境中培养至对数中期，随后分别用该七肽或仅含溶剂的对照处理，继续孵育30分钟后收集样本用于RNA制备。
本实验在两种培养基中重复开展：托德·休伊特培养基（Todd Hewitt，简称TH；Difco品牌），以及化学成分限定的FMC培养基（引自Terleckyj B, Willett NP, Shockman GD. 1975. 数种口腔致龋链球菌菌株在化学成分限定培养基中的生长. 感染与免疫（Infect Immun）. 11(4):649–655）。
TH培养基中的生物学重复设置：1组对照样本、1组处理样本，每块芯片设置6次重复；化学成分限定培养基中的生物学重复设置：2组对照样本、2组处理样本，每块芯片设置6次重复。所有样本组均完成了染料互换实验。
