Profilin 1 deficiency drives copy number alterations in MC3T3 cells

Profilin 1 –encoded by PFN1– is a small actin-binding protein with a tumour suppressive role in various adenocarcinomas and pagetic osteosarcomasd. Fix and live cell imaging showed that Profilin 1 inactivation results in multiple mitotic defects. Low-pass whole-genome sequencing analysis on eight Pfn1-KO MC3T3 cells highlighted frequent copy number alterations involving multiple chromosomes.

前纤维蛋白1（Profilin 1）由PFN1基因编码，是一种小型肌动蛋白结合蛋白，在多种腺癌及佩吉特病性骨肉瘤中具有抑癌功能。固定及活细胞成像实验结果表明，前纤维蛋白1失活会引发多种有丝分裂缺陷。对8株Pfn1基因敲除的MC3T3细胞进行低覆盖度全基因组测序分析后发现，其频繁出现涉及多条染色体的拷贝数变异。
