To understand the processes regulated by Smad4 in reactive cholangiocytes, Ctl and Smad4del mice were provided DDC diet for 2 weeks after which reactive cholangiocytes were isolated from digested injured mouse liver via FACS followed by RNA amplification and sequencing analysis. Gene set enrichment analysis of Smad4-perturbed reactive cholangiocytes implicates Smad4 in proliferative, metabolic, and inflammatory pathways. 为探究Smad4在反应性胆管上皮细胞(reactive cholangiocytes)中调控的生物学过程,本研究为对照组(Ctl)与Smad4敲除(Smad4del)小鼠提供DDC饮食造模2周;随后通过流式细胞术(FACS)从消化后的受损小鼠肝脏中分离反应性胆管上皮细胞,继而开展RNA扩增与测序分析。对经Smad4扰动的反应性胆管上皮细胞进行基因集富集分析(Gene Set Enrichment Analysis)结果表明,Smad4参与增殖、代谢及炎症通路的调控。本研究针对携带与缺失Smad4的反应性胆管上皮细胞的RNA测序(RNA-seq)数据开展比较基因表达谱分析。
