This dataset contains all original experimental data supporting the findings presented in Chapter 3 and Chapter 4 of my PhD thesis. The overall project focuses on the development of fibroblast membrane-coated nanoparticles for tumor-targeted therapy via ferroptosis induction (Chapter 3) and immunomodulation through STING pathway activation (Chapter 4). All experiments were conducted using standard in vitro assays and multiple murine tumor models, and results were recorded using a combination of imaging, molecular analysis, and functional evaluations.The uploaded dataset is organized into two main folders—Dataset (Chapter 3) and Dataset (Chapter 4)—each containing several subfolders corresponding to major experimental categories:<b>Dataset (Chapter 3)</b> includes five subfolders:<b>Function characterization</b>: TEM images, immunofluorescence images, cellular uptake and transwell migration assays (TIF format), and CSV files of nanoparticle size/zeta potential analysis using GraphPad.<b>Targeting evaluation</b>: In vitro and in vivo targeting data, including tumor spheroid uptake images and IVIS fluorescence imaging (TIF format).<b>Photothermal evaluation</b>: Thermal images (PNG/JPG) and CSV data from UV-vis spectrum analysis.<b>In vitro therapeutic efficacy</b>: Confocal microscopy images of FerroOrange staining (TIF format), and CSV files of cell viability, ELISA, BODIPY staining, and GSH analysis.<b>In vivo therapeutic efficacy</b>: H&amp;E and immunohistochemistry images (TIF/JPG), and CSV data of mouse body weight, tumor weight, and tumor MDA levels.<b>Dataset (Chapter 4)</b> includes three subfolders:<b>Function characterization</b>: Cellular uptake images (TIF) and western blot data (PDF).<b>Targeting evaluation</b>: Tumor spheroid uptake images (TIF), and fluorescence/immunofluorescence imaging results (PDF).<b>In vivo therapeutic efficacy</b>: Therapeutic outcomes and survival curves (PDF), along with CSV files of body weight, tumor volume, tumor weight, and flow cytometry data.All image formats (TIF, JPG, PNG, PDF) are directly readable and organized by experiment type. CSV files include raw and processed numerical data analyzed primarily using GraphPad Prism. No sensitive or personally identifiable information is included.This dataset provides a comprehensive and reproducible record of the experimental work conducted during my doctoral studies.

本数据集包含支撑本人博士论文第3章与第4章所呈现研究发现的全部原始实验数据。本研究整体聚焦两类方向:第3章为开发成纤维细胞膜包被纳米颗粒,通过诱导铁死亡(ferroptosis)实现肿瘤靶向治疗;第4章为通过激活STING通路(STING pathway)实现免疫调节。所有实验均采用标准体外实验方法与多种小鼠肿瘤模型开展,实验结果通过成像、分子分析及功能评估等多种手段联合记录。 本次上传的数据集分为两大主文件夹——「第3章数据集」与「第4章数据集」,每个主文件夹下均设有多个对应核心实验类别的子文件夹: ### 第3章数据集 包含5个子文件夹: 1. **功能表征**:透射电子显微镜(TEM)图像、免疫荧光图像、细胞摄取与Transwell迁移实验结果(均为TIF格式),以及采用GraphPad软件分析得到的纳米颗粒粒径/ζ电位数据CSV文件。 2. **靶向评估**:体外与体内靶向实验数据,包括肿瘤球体摄取图像与活体成像(IVIS)荧光图像(TIF格式)。 3. **光热评估**:热成像图像(PNG/JPG格式)以及紫外-可见光谱分析得到的CSV数据文件。 4. **体外治疗效能**:铁橙(FerroOrange)染色共聚焦显微镜图像(TIF格式),以及细胞活力、酶联免疫吸附实验(ELISA)、BODIPY染色与谷胱甘肽(GSH)分析的CSV数据文件。 5. **体内治疗效能**:苏木精-伊红(H&E)与免疫组化图像(TIF/JPG格式),以及小鼠体重、肿瘤重量与肿瘤丙二醛(MDA)水平的CSV数据文件。 ### 第4章数据集 包含3个子文件夹: 1. **功能表征**:细胞摄取图像(TIF格式)与蛋白质印迹(western blot)实验数据(PDF格式)。 2. **靶向评估**:肿瘤球体摄取图像(TIF格式)以及荧光/免疫荧光成像结果(PDF格式)。 3. **体内治疗效能**:治疗效果与生存曲线(PDF格式),以及体重、肿瘤体积、肿瘤重量与流式细胞术数据的CSV文件。 所有图像文件(TIF、JPG、PNG、PDF格式)均可直接读取,并按实验类型分类整理;CSV文件包含原始与处理后的数值数据,主要采用GraphPad Prism软件进行分析。本数据集未包含任何敏感信息或个人可识别信息。本数据集完整且可复现地记录了本人博士在读期间开展的全部实验工作。
