The aim of this experiment was to decipher the role of retinoic acid receptor (RAR) on interleukin(IL)-17-producing gamma delta (gdT17) T cells. Therefore we generated mice where RAR activity is defective specifically in cells expressing the transcription factor RORgt, which is constitutively on in gdT17 cells. From these mice and their littermate controls we FACS-sorted the two major gdT17 populations from the lymph node (Vg4+ and Vg4-) and subjected them to RNA-seq.
本实验旨在阐明视黄酸受体(retinoic acid receptor, RAR)对产生白细胞介素17(IL-17)的γδ T细胞(gdT17细胞)的调控作用。为此,我们构建了RAR活性在表达转录因子RORgt的细胞中特异性缺失的小鼠——该转录因子在gdT17细胞中呈组成型表达。随后,我们从该基因工程小鼠及其同窝对照小鼠的淋巴结中,通过荧光激活细胞分选(fluorescence-activated cell sorting, FACS)分离得到两种主要的gdT17细胞亚群(Vg4+与Vg4-),并对其开展RNA测序(RNA-seq)分析。
