Arbuscular mycorrhizal (AM) fungal communities were characterized using a meta-barcoding approach. The small subunit ribosomal RNA (SSU rRNA) region was amplified with the barcoded primer pair WANDA/AML2. Amplicons were sequenced using paired-end sequencing (2 × 300 bp) on the Illumina MiSeq platform. Sequence data were processed with the DADA2 denoising algorithm to generate amplicon sequence variants (ASVs). Taxonomic assignment of AM fungal sequences was performed via BLAST searches against the MaarjAM database.

本研究采用宏条形码(meta-barcoding)技术对丛枝菌根(Arbuscular Mycorrhizal, AM)真菌群落进行了特征分析。采用带条形码标记的引物对WANDA/AML2扩增核糖体RNA小亚基(SSU rRNA)区域。扩增产物通过Illumina MiSeq测序平台进行双端测序(2×300 bp)。利用DADA2去噪算法对测序数据进行处理,以生成扩增子序列变异体(ASVs)。通过针对MaarjAM数据库的BLAST比对,完成AM真菌序列的分类学注释。
