A total of 421 Upland cotton (Gossypium hirsutum) core germplasm resources were cultivated in the field in Anyang, Henan, China. Cotton boll were tagged on the day of flowering as 0 DPA (day post anthesis) samples. Three replicates were set up for each accession, fibers at 5 DPA from at least 5 cotton bolls of different plants were collected and bulked together. Fibers at 10 DPA takes the same approach to collect. A total of 38 semi-wild cotton resources were cultivated in the field in Sanya, Hainan, China. Fibers at 10 DPA were collected by the same method. RNA extraction was performed on these fiber samples followed by sequencing.
本研究于中国河南安阳大田种植421份陆地棉(Gossypium hirsutum)核心种质资源。于开花当日对棉铃进行标记,作为开花后0天(0 DPA,day post anthesis)样本;每份种质设置3次生物学重复,采集不同植株上至少5个棉铃的开花后5天(5 DPA)纤维并混合均匀,开花后10天(10 DPA)的纤维样本亦采用相同方法采集。此外,38份半野生棉种质资源于中国海南三亚大田种植,其开花后10天(10 DPA)的纤维样本采用相同方法采集。对上述全部纤维样本进行RNA提取后开展测序。
