To investigate the effects of pathological IFN-γ stimulation on neurons in the context of subsequent activation, we established cell cultures of primary mouse neurons receiving pathological priming or no priming. We then performed RNA-Seq on these cells after vehicle, physiological, and pathological IFN-γ restimulation. Overall design: Cultured primary mouse neurons were sequenced after treatment. Cells from each mouse were separated into six wells and each well given a different treatment. Each well was sequenced on two lanes and the data combined.
为探究病理性干扰素-γ(IFN-γ)预刺激对神经元后续激活状态的影响,我们构建了接受病理性预刺激或未接受预刺激的原代小鼠神经元细胞培养模型。随后分别在载体对照、生理水平及病理性干扰素-γ再刺激后,对上述细胞开展转录组测序(RNA-Seq)。 实验整体设计:对经不同处理培养的原代小鼠神经元进行测序。每只小鼠来源的细胞被均分至6个孔板,每孔接受独有的处理方案;每个孔的样本在两个测序泳道(lane)上进行测序,最终合并所有测序数据。
