As a single strand DNA binding protein, RPA1 participates in various cellular processes including DNA replication and DNA repair. However, the role of RPA1 in T cells is largely unknown. We generated a mouse model in which RPA1 was specifically deleted in T cells. Through single cell RNA sequencing, we reveal the immune landscape in RPA1 conditional knockout mice. Overall design: Study of immune cells in thymus from WT or RPA1 conditional knockout mice using 10x scRNA-seq technologies.
作为单链DNA结合蛋白(single strand DNA binding protein),RPA1参与DNA复制与DNA修复等多种细胞生物学过程。然而,RPA1在T细胞中的具体作用尚不明确。我们构建了T细胞特异性敲除RPA1的条件性敲除(conditional knockout)小鼠模型。通过单细胞RNA测序(single cell RNA sequencing),我们揭示了RPA1条件性敲除小鼠的免疫细胞图谱。实验总体设计:采用10x scRNA-seq技术,对野生型(wild type, WT)及RPA1条件性敲除小鼠的胸腺免疫细胞进行研究。
