We study gene expression of Pseudomonas aeruginosa populations directly sequencing informative RNA species obtained from fresh expectorates of cystic fibrosis (CF) patients chronically infected with P. aeruginosa. At the same time, we isolate representative clones from the very same sputum samples and sequence the genome in order to evaluate the genetic distance between the different populations.We use the data to study gene expression of P. aeruginosa in pathophysiological conditions and to define a common transcriptional program achieved in CF airways independently of the evolutionary trajectory.
本研究以慢性感染铜绿假单胞菌(Pseudomonas aeruginosa)的囊性纤维化(CF)患者的新鲜痰液样本为研究对象,通过直接测序样本中的特征性RNA分子,分析铜绿假单胞菌种群的基因表达情况。与此同时,我们从同一痰液样本中分离得到代表性克隆株,并对其基因组进行测序,以此评估不同种群间的遗传距离。本研究利用上述数据集,探究铜绿假单胞菌在病理生理状态下的基因表达特征,并明确CF患者气道中不依赖于进化轨迹的共性转录程序。
