Urinary bladder wound healing is today pooorly chracterized. MicroRNAs are small non-coding RNA molecules with regulatory functions. In this study we aimed at identifying microRNAs expressed during bladder wound healing. We performed Affymetrix microRNA profiling of the rodent urinary bladder during healing of a surgically created wound. Rat urinary bladder were collected at 0 hours ( control non wounded animals), 2 days and 8 days post insicional wound and sutures, 4 animals per time point were incliuded. The Affymetrix microRNA GeneChip Array plates (Affymetrix, Thermo Fisher) were used in this study according to the manufacturer’s instructions at the core facility for Bioinformatics and Expression Analysis (BEA, Karolinska Institutet, Stockholm, Sweden)
目前,膀胱创伤愈合的相关机制尚缺乏充分的表征。微小RNA(microRNAs)是一类具备调控功能的小型非编码RNA分子。本研究旨在筛选出膀胱创伤愈合过程中表达的微小RNA。本研究针对手术构建创伤后的啮齿类动物膀胱,采用Affymetrix平台开展微小RNA表达谱分析。本研究分别于术后0小时(未受创伤的对照组动物)、术后2天及术后8天,在手术切口创伤并缝合后收集大鼠膀胱组织,每个时间点纳入4只实验动物。本研究使用Affymetrix微小RNA基因芯片板(Affymetrix,Thermo Fisher),并严格按照制造商的操作说明,在瑞典斯德哥尔摩卡罗林斯卡学院生物信息学与表达分析核心实验室(BEA)完成全部实验操作。
